As Richard pointed out, duplicity can be very picky about how paths are 
specified. If your backup line was something like:

duplicity /home/user protocol:///some/remote/location

....and you want to restore file /home/user/Mail/trash/2324, you'd only write 
everything after the root folder:

duplicity --file-to-restore Mail/trash/2324 protocol:///some/remote/ocation 
/any/local/location
