No.  You can serve any number of repositories with a single port, just
add a --allow-root= option for each repository.

You can also do that, if you want, but there's probably no good reason
to.  You just need to make multiple entries in your /etc/inetd.conf (one
for each port) and then have the clients specify the port number to use
explicitly.
