Oops, you are right. I was wrong. Since version 0.17, gettextize *does*
support multiple --po-dir options.

autopoint does it automatically. It infers the list of po/ dirs.
Just try it.

What should I add to the docs, if a feature is automatic?

If you want to reopen this bug report, please provide a test case
(configure.ac, Makefile.am etc. with several po/ directories) and
a sequence of commands, including an autopoint invocation, that
supports your claim.
